Combination of Ursodeoxycholic Acid and Glutathione Improves Intestinal Morphology in Cholestasis by Downregulating TNF-α Expression

چکیده
BACKGROUND: Cholestasis caused by obstruction of the common bile duct and may developed gut-derived sepsis due to reactive oxygen species (ROS) accumulation. Ursodeoxycholic acid (UDCA) glutathione are widely known for their antioxidant properties, that might be beneficial against ROS. However, effects UDCA-glutathione combination ROS have not been well elucidated in previous studies. Thus, this study was conducted evaluate tumor necrosis factor (TNF)-α level height terminal ileal mucosal villus after administration cholestasis rat model.METHODS: Twenty-eight male Sprague Dawley rats were randomly grouped into four treatment groups, each group consisted seven had previously undergone ligation. Three groups received on stratified dose, while other one only UDCA. Each given 21 days. Ileal samples collected from stained with mouse anti TNF-a antibody hematoxylin-eosin (HE). Immunohistochemistry histopathological examination done using microscope then calculated ImageJ.RESULTS: The UDCA decreased TNF-α expression (p<0.05) compared group, particularly 20 mg 15 supplementation 30 (p<0.05). mucosa villous higher all three dosage variations group.CONCLUSION: downregulates improves ileum cholestasis.KEYWORDS: cholestasis, glutathione, intestinal height, TNF-α,

Intrahepatic cholestasis of pregnancy: changes in maternal-fetal bile acid balance and improvement by ursodeoxycholic acid.
Intrahepatic cholestasis of pregnancy (ICP) is a disease characterized by generalized pruritus and biochemical cholestasis that appears typically during the last trimester of gestation. The most predictive and accurate markers for diagnosis and follow-up of ICP are increased total bile acid levels (above 11,0 micromol/L), enhanced cholic acid percentage (above 42%) and decreased glycine/taurine...
متن کاملUrsodeoxycholic acid in cholestasis: linking action mechanisms to therapeutic applications.
UDCA (ursodeoxycholic acid) is the therapeutic agent most widely used for the treatment of cholestatic hepatopathies. Its use has expanded to other kinds of hepatic diseases, and even to extrahepatic ones. Such versatility is the result of its multiple mechanisms of action. UDCA stabilizes plasma membranes against cytolysis by tensioactive bile acids accumulated in cholestasis. UDCA also halts ...

15 صفحه اولBile acid patterns in meconium are influenced by cholestasis of pregnancy and not altered by ursodeoxycholic acid treatment.
BACKGROUND Data on meconium bile acid composition in newborn babies of patients with intrahepatic cholestasis of pregnancy (ICP) are relatively scant, and changes that occur on ursodeoxycholic acid (UDCA) administration have not been evaluated. AIMS To investigate bile acid profiles in meconium of neonates from untreated and UDCA treated patients with ICP. Maternal serum bile acid composition...
